Short Description
The BioDeviceLab Human Lipopolysaccharide-Binding Protein (LBP) ELISA Kit is a high-sensitivity sandwich immunoassay designed for quantitative measurement of LBP in serum, plasma, and cell culture supernatants. The assay enables accurate assessment of microbial translocation, endotoxin exposure, and epithelial barrier dysfunction and is optimized for gut-on-chip validation and host–microbiome interaction studies.

Product Overview
The BioDeviceLab Human LBP ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of lipopolysaccharide-binding protein (LBP), an acute-phase protein primarily produced by hepatocytes and intestinal epithelial cells. LBP plays a central role in innate immune sensing by binding bacterial lipopolysaccharide (LPS) and facilitating its presentation to CD14/TLR4 signaling complexes.

In gastrointestinal systems, circulating and locally secreted LBP serves as a sensitive biomarker of microbial translocation, epithelial barrier disruption, and dysregulated host–microbiome interactions. Elevated LBP levels are associated with increased gut permeability, systemic inflammation, metabolic dysregulation, and inflammatory bowel disease. In gut-on-chip platforms, LBP provides a robust quantitative readout of endotoxin exposure, epithelial leakage, and immune activation under microbial challenge. This assay combines strong analytical sensitivity, high specificity, and excellent reproducibility, making it suitable for mechanistic studies and advanced biodevice validation workflows.

Intended Use
This kit is intended for the quantitative determination of LBP in:
• Serum
• Plasma
• Cell culture supernatants

Applications include gut-on-chip permeability and microbial translocation studies, host–microbiome interaction analysis, endotoxin challenge models, inflammatory signaling research, drug and biologic screening, and biodevice calibration and benchmarking.

Assay Principle
This assay employs a sandwich ELISA format:
• LBP present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to LBP binds to the captured protein.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the LBP concentration in the sample.

Calibration Curve
• A calibration curve is generated using recombinant human LBP standards with known concentrations.
• Mean absorbance values are plotted against LBP concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.

Analytical Performance
Dynamic Detection Range
• 0.5 – 50 µg/mL

Analytical Sensitivity
• Limit of Detection (LOD): ≤ 0.2 µg/mL

Precision
• Intra-assay CV: < 9%
• Inter-assay CV: < 7%

Selectivity & Specificity
The assay demonstrates high specificity for human LBP with minimal cross-reactivity to related acute-phase proteins (CRP, CD14, MD-2) (< 1%).
