The BioDeviceLab Human Interleukin-10 (IL-10) ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of IL-10 in biological samples. IL-10 is a key anti-inflammatory cytokine produced by immune cells and epithelial cells that plays a central role in limiting excessive inflammatory responses and maintaining intestinal immune homeostasis.

IL-10 suppresses the production of pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6, promotes epithelial barrier recovery, and regulates immune tolerance to commensal microbiota. Dysregulation of IL-10 signaling is strongly associated with inflammatory bowel disease, chronic intestinal inflammation, and impaired mucosal healing. In gut-on-chip platforms, IL-10 serves as a critical biomarker for evaluating immune resolution, epithelial recovery, and therapeutic efficacy of anti-inflammatory or immunomodulatory interventions. Assay Principle
This assay employs a sandwich ELISA format:
• IL-10 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to IL-10 binds to the captured protein.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the IL-10 concentration in the sample.

Calibration Curve
• A calibration curve is generated using recombinant human IL-10 standards with known concentrations.
• Mean absorbance values are plotted against IL-10 concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.

Limit of detection (LOD): ≤ 2 pg/mL
Intra-assay CV: < 10%

Selectivity & Specificity
