Product Description
The BioDeviceLab Human Vascular Endothelial Growth Factor (VEGF) ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of VEGF in biological samples. VEGF is a central regulator of angiogenesis, vascular permeability, and endothelial cell survival and plays a critical role in tissue remodeling, inflammation-driven vascular responses, and epithelial–vascular crosstalk.

In gut-on-chip and intestinal disease models, VEGF is a key biomarker linking epithelial stress, hypoxia, inflammation, and microvascular remodeling. Elevated VEGF levels are associated with inflammatory bowel disease, mucosal injury, tumor angiogenesis, and barrier dysfunction, while controlled VEGF signaling is essential for tissue repair and regeneration. This assay provides high analytical sensitivity, strong specificity, and robust reproducibility, making it suitable for mechanistic angiogenesis studies, drug screening, and advanced biodevice validation workflows.

Intended Use
This kit is intended for the quantitative determination of human VEGF in:
• Serum
• Plasma
• Cell culture supernatants
• Tissue culture media

Applications include gut-on-chip vascularization studies, angiogenesis research, inflammation-driven vascular remodeling, drug and biologic screening, and biodevice calibration and benchmarking.

Assay Principle
This assay employs a sandwich ELISA format:
• VEGF present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to VEGF binds to the captured protein.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the VEGF concentration in the sample.

Calibration Curve
• A calibration curve is generated using recombinant human VEGF standards with known concentrations.
• Mean absorbance values are plotted against VEGF concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.

Analytical Performance
Dynamic detection range: 10 – 1,000 pg/mL
Limit of detection (LOD): ≤ 5 pg/mL
Intra-assay CV: < 8%
Inter-assay CV: < 8%

Selectivity & Specificity
The assay demonstrates high specificity for human VEGF with minimal cross-reactivity to related growth factors and cytokines (< 1%).
