The BioDeviceLab Human Mucin-2 (MUC2) ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of MUC2 in biological samples. MUC2 is the dominant gel-forming mucin secreted by goblet cells and constitutes the primary structural component of the intestinal mucus layer.

The mucus layer plays a critical role in separating luminal microbes from the epithelial surface, regulating microbial colonization, and maintaining intestinal homeostasis. Alterations in MUC2 production, secretion, or structure are associated with inflammatory bowel disease, infection, dysbiosis, and barrier dysfunction. In gut-on-chip platforms, MUC2 serves as a key functional biomarker for evaluating mucus layer formation, goblet cell function, and microbial–epithelial interactions under physiological and pathological conditions. This assay combines high analytical sensitivity, strong specificity, and robust reproducibility, making it suitable for both mechanistic studies and advanced biodevice validation workflows.

Assay Principle
This assay employs a sandwich ELISA format:
• MUC2 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to MUC2 binds to the captured protein.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the MUC2 concentration in the sample.

Calibration Curve
• A calibration curve is generated using recombinant human MUC2 standards with known concentrations.
• Mean absorbance values are plotted against MUC2 concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.

Limit of detection (LOD): ≤ 0.05 ng/mL
Intra-assay CV: < 10%
Inter-assay CV: < 12%

Selectivity & Specificity
The assay demonstrates high specificity for MUC2 with minimal cross-reactivity to other mucins (MUC1, MUC4, MUC5AC) or unrelated glycoproteins (< 1%).
