Product Overview
The BioDeviceLab Human IL-22 ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of IL-22, a key cytokine produced by Th17/Th22 cells and innate lymphoid cells that acts primarily on epithelial tissues. IL-22 supports mucosal barrier maintenance by promoting epithelial regeneration, enhancing tight junction function, and inducing antimicrobial peptides.

In the gastrointestinal tract, IL-22 is a central mediator of epithelial repair and host defense during infection, inflammation, and barrier disruption. Dysregulated IL-22 signaling is implicated in inflammatory bowel disease, infection-driven epithelial injury, and altered host–microbiome interactions. In gut-on-chip models, IL-22 provides a strong readout for epithelial restoration programs, antimicrobial induction pathways, and evaluation of therapeutics that modulate mucosal immunity. This assay combines high analytical sensitivity, strong specificity, and robust reproducibility, making it suitable for mechanistic studies and advanced biodevice validation workflows.

Intended Use
This kit is intended for the quantitative determination of IL-22 in:
• Serum
• Plasma
• Cell culture supernatants

Applications include gut-on-chip epithelial repair studies, mucosal immunity research, host–microbiome interaction analysis, infection and inflammation models, drug and biologic screening, and biodevice calibration and benchmarking.

Assay Principle
This assay employs a sandwich ELISA format:
• IL-22 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to IL-22 binds to the captured cytokine.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the IL-22 concentration in the sample.

Calibration Curve
• A calibration curve is generated using recombinant human IL-22 standards with known concentrations.
• Mean absorbance values are plotted against IL-22 concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.

Analytical Performance
Dynamic Detection Range
• 5 – 1,000 pg/mL

Analytical Sensitivity
• Limit of Detection (LOD): ≤ 4 pg/mL

Precision
• Intra-assay CV: < 10%
• Inter-assay CV: < 9%

Selectivity & Specificity
The assay demonstrates high specificity for IL-22 with minimal cross-reactivity to related cytokines (IL-17A, IL-10, IL-6, IL-26) (< 1%).
