The BioDeviceLab Human Interleukin-8 (IL-8 / CXCL8) ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of IL-8 in biological samples. IL-8 is a pro-inflammatory chemokine secreted by intestinal epithelial cells, immune cells, and endothelial cells in response to microbial products, cytokines, and cellular stress.

IL-8 plays a central role in neutrophil recruitment, epithelial immune signaling, and amplification of inflammatory responses in the gut. Elevated IL-8 levels are observed in inflammatory bowel disease, infection, dysbiosis, and epithelial barrier disruption. In gut-on-chip platforms, IL-8 serves as a robust and dynamic biomarker for quantifying epithelial inflammatory activation under microbial challenge, cytokine stimulation, drug exposure, or barrier injury. This assay combines high analytical sensitivity, strong specificity, and robust reproducibility, making it suitable for both mechanistic inflammation studies and advanced biodevice validation workflows.

Assay Principle
This assay employs a sandwich ELISA format:
• IL-8 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to IL-8 binds to the captured protein.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the IL-8 concentration in the sample.

Calibration Curve
• A calibration curve is generated using recombinant human IL-8 standards with known concentrations.
• Mean absorbance values are plotted against IL-8 concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.

Limit of detection (LOD): ≤ 2 pg/mL
Intra-assay CV: < 10%
Inter-assay CV: < 8%

Selectivity & Specificity
The assay demonstrates high specificity for IL-8 with minimal cross-reactivity to other cytokines and chemokines (IL-6, IL-1β, TNF-α, MCP-1) (< 1%).
