Short Description
The BioDeviceLab Human Interleukin-22 (IL-22) ELISA Kit is a high-sensitivity sandwich immunoassay for quantitative measurement of IL-22 in serum, plasma, and cell culture supernatants. The assay enables precise assessment of epithelial barrier regulation, mucosal immunity, and immune–epithelial crosstalk in gut-on-chip systems.

For Research Use Only (RUO).

Product Overview
The BioDeviceLab Human IL-22 ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed to quantify IL-22, a key cytokine produced by Th17 cells, innate lymphoid cells (ILC3s), and NK cells. IL-22 acts primarily on epithelial cells, where it regulates barrier integrity, antimicrobial peptide production, epithelial regeneration, and wound healing.

In gastrointestinal biology, IL-22 is a central mediator of host defense and tissue repair at the mucosal surface. Dysregulated IL-22 signaling is implicated in inflammatory bowel disease, infection-driven epithelial injury, and altered host–microbiome interactions. In gut-on-chip platforms, IL-22 serves as a critical biomarker for epithelial resilience, immune-driven regeneration, and evaluation of immunomodulatory or microbiome-targeted therapies. This assay combines high analytical sensitivity, strong specificity, and reproducible performance suitable for mechanistic studies and advanced biodevice validation workflows.

Intended Use
This kit is intended for the quantitative determination of IL-22 in:
• Serum
• Plasma
• Cell culture supernatants

Applications include gut-on-chip epithelial barrier studies, Th17 and ILC3 immune profiling, host–microbiome interaction research, inflammation and infection models, drug and biologic screening, and biodevice calibration and benchmarking.

Assay Principle
This assay employs a sandwich ELISA format:
• IL-22 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to IL-22 binds to the captured cytokine.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to IL-22 concentration.

Calibration Curve
• Generated using recombinant human IL-22 standards.
• Mean absorbance plotted versus concentration on a semi-logarithmic scale.
• Four-parameter logistic (4PL) regression recommended.
• Sample concentrations interpolated from the standard curve.

Analytical Performance
Dynamic Detection Range
• 5 – 2,000 pg/mL

Analytical Sensitivity
• Limit of Detection (LOD): ≤ 2 pg/mL

Precision
• Intra-assay CV: < 8%
• Inter-assay CV: < 8%

Selectivity & Specificity
High specificity for human IL-22 with minimal cross-reactivity to IL-17A, IL-10, IL-20 family cytokines, and IFN-γ (< 1%).
