Product Overview
The BioDeviceLab Human Brain-Derived Neurotrophic Factor (BDNF) ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of BDNF, a key neurotrophin that regulates neuronal survival, differentiation, synaptic plasticity, and learning-related signaling. BDNF is produced by neurons and glial cells and can also be modulated by immune activity, systemic inflammation, and microbial metabolites.
In neuroinflammation and gut–brain axis research, BDNF serves as a functional biomarker reflecting neurotrophic support and adaptive responses to stressors. Reduced BDNF is associated with neurodegenerative processes, chronic inflammation, and impaired neuronal resilience, while recovery of BDNF is frequently used as a positive endpoint in therapeutic studies. In gut-on-chip and neuro-on-chip platforms, BDNF provides a quantitative readout of barrier-mediated neurotoxicity, inflammatory crosstalk, and intervention efficacy. This assay combines high analytical sensitivity, strong specificity, and reproducible performance suitable for mechanistic studies, drug screening, and biodevice validation workflows.

Assay Principle
This assay employs a sandwich ELISA format:
• BDNF present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to BDNF binds to the captured protein.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to BDNF concentration.
Calibration Curve
• Generated using recombinant human BDNF standards with known concentrations.
• Mean absorbance values plotted versus BDNF concentration on a semi-logarithmic scale.
• Four-parameter logistic (4PL) regression recommended.
• Sample concentrations interpolated from the standard curve.
