Product Overview
The BioDeviceLab Human IL-6 ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of IL-6, a pleiotropic cytokine produced by epithelial cells, macrophages, fibroblasts, and immune cells in response to infection, tissue damage, and microbial stimulation. IL-6 plays a central role in acute-phase responses, immune cell recruitment, epithelial barrier modulation, and chronic inflammation.
In gastrointestinal systems, IL-6 is a critical biomarker of epithelial stress, inflammatory activation, and dysregulated host–microbiome interactions. Elevated IL-6 levels are associated with inflammatory bowel disease, infection-driven epithelial injury, and impaired barrier function. In gut-on-chip platforms, IL-6 provides a robust quantitative readout of immune activation, epithelial–immune communication, and therapeutic efficacy. This assay combines high analytical sensitivity, excellent specificity, and reproducible performance, making it suitable for both mechanistic studies and advanced biodevice validation workflows.
Intended Use
This kit is intended for the quantitative determination of IL-6 in:
• Serum
• Plasma
• Cell culture supernatants
Applications include gut-on-chip inflammation modeling, host–microbiome interaction studies, cytokine profiling, infection and injury models, drug and biologic screening, and biodevice calibration and benchmarking.
Assay Principle
This assay employs a sandwich ELISA format:
• IL-6 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to IL-6 binds to the captured cytokine.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a colorimetric signal measured at 450 nm.
• Absorbance is directly proportional to the IL-6 concentration in the sample.
Calibration Curve
• A calibration curve is generated using recombinant human IL-6 standards with known concentrations.
• Mean absorbance values are plotted against IL-6 concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.
Analytical Performance
Dynamic Detection Range
• 2 – 1,000 pg/mL
Analytical Sensitivity
• Limit of Detection (LOD): ≤ 1 pg/mL
Precision
• Intra-assay CV: < 8%
• Inter-assay CV: < 8%
Selectivity & Specificity
The assay demonstrates high specificity for IL-6 with minimal cross-reactivity to related cytokines (IL-1β, IL-8, TNF-α, IL-10) (< 1%).
