Product Overview
The BioDeviceLab Human MCP-1 / CCL2 ELISA Kit is a high-performance enzyme-linked immunosorbent assay developed for the quantitative determination of MCP-1, a key chemokine involved in monocyte, macrophage, and T-cell recruitment to sites of inflammation. MCP-1 is secreted by epithelial cells, endothelial cells, fibroblasts, and immune cells in response to inflammatory stimuli and microbial signals.
In the gastrointestinal tract, MCP-1 plays a central role in shaping immune-cell trafficking, epithelial–immune crosstalk, and chronic inflammatory responses. Dysregulated MCP-1 signaling is associated with inflammatory bowel disease, infection-driven inflammation, fibrosis, and impaired barrier recovery. In gut-on-chip platforms, MCP-1 serves as a critical biomarker for immune recruitment gradients, chemotactic signaling, and evaluation of immunomodulatory therapeutics. This assay combines high sensitivity, excellent specificity, and reproducible performance, making it suitable for both mechanistic studies and advanced biodevice workflows.
Intended Use
This kit is intended for the quantitative determination of MCP-1 / CCL2 in:
• Serum
• Plasma
• Cell culture supernatants
Applications include gut-on-chip immune recruitment studies, host–microbiome interaction research, inflammatory signaling analysis, drug and biologic screening, and biodevice calibration and benchmarking.
Assay Principle
This assay employs a sandwich ELISA format:
• MCP-1 present in the sample binds to capture antibodies immobilized on a 96-well microplate.
• A biotinylated detection antibody specific to MCP-1 binds to the captured chemokine.
• An enzyme conjugate is added, followed by chromogenic substrate (TMB).
• The enzymatic reaction generates a color signal measured at 450 nm.
• Absorbance is directly proportional to the MCP-1 concentration in the sample.
Calibration Curve
• A calibration curve is generated using recombinant human MCP-1 standards with known concentrations.
• Mean absorbance values are plotted against MCP-1 concentration on a semi-logarithmic scale.
• A four-parameter logistic (4PL) regression model is recommended for curve fitting.
• Sample concentrations are calculated by interpolation from the standard curve.
Analytical Performance
Dynamic Detection Range
• 10 – 1,000 pg/mL
Analytical Sensitivity
• Limit of Detection (LOD): ≤ 3 pg/mL
Precision
• Intra-assay CV: < 8%
• Inter-assay CV: < 7%
Selectivity & Specificity
The assay demonstrates high specificity for MCP-1 with minimal cross-reactivity to related chemokines and cytokines.
Representative cross-reactivity:
• MCP-1 / CCL2: 100%
• IL-8, CXCL10, CCL5, TNF-α: < 1%
